Vektor Facility

Um Erbgut gezielt in Zellen zu schleusen und die genetischen Ursachen von Krankheiten im Labor zu untersuchen, bieten sich Viruspartikel als Gen-Fähren an. Dabei werden virale Gensequenzen durch die Fremd-DNA von Interesse ersetzt und in das virale Capsid eingepackt. Die viralen Hüllproteine bestimmen dabei den Serotyp und auch die Spezifität für unterschiedliche Gewebe bzw. Zellen. Die Replikationsmaschinerie der rekombinanten Viruspartikel ist zur Gewährleistung der größtmöglichen Sicherheit für die/den Anwender/in so modifiziert, dass lediglich eine einmalige Transduktion der Zielzelle zur Einschleusung des Transgens erfolgt, so dass das Gen in der Zelle ausgepackt und aktiviert wird, aber keine Vermehrung von Viren erfolgen kann.

Rekombinante Viruspartikel können für Funktionsstudien genutzt werden, in deren Rahmen entweder ein "knock-in" (bzw. "rescue" eines knock-outs) durch rekombinante Expression eines Transgens vermittelt wird, oder aber ein endogenes Protein durch RNA-Interferenz z.B. mittels short interfering RNA (shRNA) herunter reguliert wird.

Die Vektor Core Unit, die wissenschaftlich mit dem Institut für Experimentelle Pharmakologie und Toxikologie assoziiert ist, unterstützt Wissenschaftler/innen bei der Auswahl und der Etablierung des geeigneten Virussystems für ihre Forschung. Je nach Anforderung an die Größe des zu verpackenden Gens, die Zell- bzw. Gewebespezifität des Transfers in vitro oder in vivo bieten sich unterschiedliche Virustypen an. Darüber hinaus muss die Stabilität und Persistenz der Genexpression sowie die erforderlichen Virusmengen mit dem Umfang und der Dauer der durchzuführenden Studie abgestimmt werden.

Serviceleistungen

Nicht jedes Virus ist in der Lage, unterschiedliche Zelltypen mit gleicher Effizienz zu infizieren: Tropismus, Infektiösität, Persistenz in der Zielzelle und Toxizität unterscheiden sich mitunter signifikant. Oftmals ist eine erfolgreiche Infektion in vitro nicht auf in vivo-Studien übertragbar. Der Einsatz eines Gentransfervektors richtet sich daher stark nach dem avisierten Projektziel. Sollten Literaturrecherchen keinen Anhaltspunkt für die Suszeptibilität Ihrer Zielzellen geben, so steht in der Vektor Facility eine Auswahl an Virusstocks mit Reportergenen hierfür zur Verfügung (siehe PDF unten).

Die Vektor Facility bietet die Produktion viraler Gentransfervektoren individuell nach Kundenwunsch mit Hilfe entsprechender Transferplasmide an. Die Wahl des optimalen Transferplasmids ist u.a. von der Größe des Transgens, des Promoters und der gewünschten Expressionsdauer abhängig. In der Vektor Facility steht eine Auswahl an Transferplasmiden zur Verfügung, die wir Ihnen zur Insertion Ihres gene of interest anbieten können. Bei der Auswahl eines geeigneten Vehikels stehen wir Ihnen gerne für Diskussionen zur Verfügung. Folgende rekombinante Viren können derzeit in der Vektor Facility produziert werden:
