Regulation von Entzündung und Regeneration der Leber

Die Leber hat ein einzigartiges Potential zur Regeneration selbst schwerer Verletzungen. Leberentzündung ist eine physiologische Antwort auf Verletzungen, die zur Heilung und Regeneration des Organs nötig ist. Viele Lebererkrankungen sind jedoch durch chronische Leberschädigung und chronische Entzündung gekennzeichnet, was häufig zu aberranter Regeneration in Form von Fibrose, Zirrhose oder Lebertumoren führt. Das Projekt zielt darauf, die Zellen und Moleküle zu identifizieren, die Entzündung und Regeneration in der Leber beeinflussen.

Verschiedene Leberzelltypen, wie Hepatozyten, Kupfferzellen, Dendritische Zellen der Leber oder Sinusoidale Endothelzellen können T Zell-Antworten gegen Leberantigene oder Hepatitisvirus auslösen oder modulieren. Wir untersuchen die Auswirkungen der Immunstimulation durch die verschiedenen Leberzellen in Bezug auf T Zell-Differenzierung, entzündliche Aktivitätentzündliche Stimulation oder Suppression.
Die Relevanz dieser Befunde wird in vivo in Mausmodellen einer Virushepatitis (Lymphozytäres Choriomeningitis-Virus) bzw. von Autoimmunerkrankung (Modellen der autoimmunen Hepatitis, experimentellen Cholangitis sowie der experimentellen autoimmunen Enzephalomyelitis) untersucht.
Die Rolle von Zytokinen bei Entzündung und Regeneration der Leber wird mit Hilfe transgener Mäuse untersucht, die in der Leber spezifische Zytokine exprimieren.

Ein weiterer Schwerpunkt des Labors besteht in der Analyse der Immunregulation mit besonderem Schwerpunkt auf derdie Rolle von regulatorischen Zellpopulationen und Zytokinen in verschiedenen experimentellen Entzündungsmodellen.

Die Erfahrungen aus den experimentellen Arbeiten führen zunehmend zur Untersuchung der Mechanismen der Entzündung, Regulation und Regeneration an humanen Proben.
